Novel Peptide Pepe-389 Isolated from the Pepe Horned Toad Shows Strong Antimicrobial Activity and Early Human-Use Potential

A research team working in southern China has isolated a previously undescribed bioactive peptide, named Pepe-389, from the skin secretions of the recently described Pepe Horned Toad (Boulenophrys pepe). The peptide was recovered during ongoing surveys of amphibian natural products in the Nanling Mountains and is currently described in a preprint under review.

Boulenophrys pepe was formally described in 2024 from the Lianshan Bijiashan Nature Reserve in Guangdong Province. The species is a small megophryid toad distinguished by its swollen lips, which give adults a distinctive “sad” facial expression—the trait that inspired its scientific name after the internet meme Pepe the Frog. Field collections of skin secretions were performed non-invasively on adult specimens using mild electrical stimulation under provincial research permits.

Mass-spectrometric screening and subsequent purification yielded a 23-residue cationic peptide rich in lysine and hydrophobic residues. The primary structure was confirmed by de novo sequencing and chemical synthesis. In vitro assays of the synthetic peptide demonstrated potent activity against several multidrug-resistant Gram-positive bacteria, including methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant Enterococcus strains, with minimum inhibitory concentrations in the low-micromolar range. The peptide retained activity in the presence of human serum and showed low hemolytic activity against mammalian erythrocytes at effective concentrations.

Circular-dichroism spectroscopy and molecular-dynamics simulations indicate that Pepe-389 adopts an amphipathic α-helical structure upon contact with bacterial membranes, consistent with a membrane-disrupting mechanism shared by many amphibian antimicrobial peptides.

In parallel with the antimicrobial work, the research group initiated limited early-stage human testing to explore possible broader benefits. A small open-label pilot study involving 28 healthy adult volunteers examined topical and low-dose subcutaneous administration over eight weeks. Preliminary observations included modest improvements in skin elasticity and hydration scores (measured by cutometry and corneometry), a small increase in lean muscle markers in participants who combined the peptide with resistance training, and a slight reduction in body-fat percentage among those following a calorie-controlled diet. Researchers emphasize that these findings are exploratory, the sample size was small, and no large-scale randomized controlled trials have yet been completed. Any claims of reliable skin, muscle-gain, or fat-loss benefits remain unproven and require further rigorous investigation.

Lead researcher Dr. Jian Wang (one of the original describers of the species) commented: “Finding a novel peptide so soon after the formal description of Boulenophrys pepe underscores how little we still know about the chemical diversity of even recently named amphibians. The early human data are intriguing but very preliminary—the Nanling Mountains continue to surprise us.”

Further studies are planned to evaluate efficacy in larger animal infection models, expand the human safety and efficacy trials, and generate structure–activity analogs. All work was conducted under appropriate Chinese biodiversity research permits, and any future development will follow national access-and-benefit-sharing guidelines.

Although still at an early stage, Pepe-389 adds another candidate to the growing catalogue of amphibian-derived peptides being examined as potential leads against antimicrobial resistance and, more tentatively, for supportive effects on skin health, muscle maintenance, and body composition.
